AAV Affinity CIP1 Solution is an acidic cleaning-in-place reagent for chromatography workflows involving recombinant adeno-associated virus (AAV). It is used to clean AAV affinity columns and system flow paths after product elution by removing process-derived impurities such as host cell proteins, nucleic acids, lipids, and residual feed components. It is applicable to affinity resins designed for common AAV serotypes and related engineered capsids.

The solution is intended for use according to column and system manufacturer instructions for contact time, linear velocity, and temperature, followed by thorough flushing and re-equilibration with the designated binding buffer. Store in the original container under standard laboratory chemical storage conditions, and handle using appropriate personal protective equipment and corrosion-resistant materials.

  • 0.1M Phosphoric Acid
  • Adjusted to pH 2.0
  • AAV Affinity Column Cleaning-in-Place (CIP): Acidic CIP to strip tightly bound contaminants from AAV affinity resins; aligns with 0.1 M phosphoric acid-based cleaning.

  • Resin Regeneration Between Cycles: Restores binding capacity of AAV affinity media for consistent performance across multiple purification runs.

  • Impurity Clearance: Removes host cell proteins, nucleic acids (DNA/RNA), and lipids from the column following AAV elution.

  • Endotoxin Reduction: Lowers endotoxin burden from Escherichia coli-derived feedstocks on affinity resins and flow paths.

  • Sanitization/Bioburden Control: Acidic sanitization of chromatography hardware and media to control bioburden between batches.

  • Process Maintenance in GMP Workflows: Supports validated cleaning procedures for AAV purification skids and columns.

  • pH: 2.00 ± 0.10
