AAV Affinity Wash Buffer is a process buffer used in affinity chromatography workflows for adeno-associated virus (AAV) purification. It is applied after sample loading to remove unbound and non-specifically associated host cell proteins, nucleic acids, and other impurities while maintaining AAV capsid interaction with the affinity ligand prior to elution.

This buffer is suitable for wash steps when purifying AAV from clarified cell lysates or culture supernatants produced in common expression systems, and can be used across multiple AAV capsid serotypes in ligand-based affinity capture processes.

  • 20mM Tris Base
  • 1.0M Sodium Chloride
  • Adjusted to pH 7.5
  • Affinity Capture Wash: Removes nonspecific host proteins and nucleic acids from AAV-loaded affinity resins prior to elution.

  • Column Equilibration and Post-Load Wash: Used to equilibrate columns and perform consistent wash steps in AAV purification workflows.

  • Harvest/Lysate Clarification Wash: Applied during purification of AAV from HEK293 or Sf9 harvests to reduce process contaminants.

  • Intermediate Stringency Wash: Minimizes carryover of empty capsids and process-related impurities before polishing.

  • Serotype-Agnostic Wash: Supports wash steps across multiple AAV serotypes (e.g., AAV1–AAV9, rh10, DJ, Anc80) on common affinity resins.

  • FPLC/AKTA-Compatible Washing: Provides stable pH control during automated wash steps on chromatography systems.

  • Pre-Polishing Preparation: Conditions captured AAV for downstream buffer exchange and polishing chromatography.

  • Consistency in RUO Workflows: Delivers reproducible wash performance in research-use-only AAV purification protocols.
