AAV Affinity Elution Buffer is an acidic buffer formulated for the elution step in affinity chromatography workflows for adeno-associated virus (AAV) capsids. It is intended to release AAV particles from affinity media that require low-pH conditions for desorption, supporting purification processes across serotypes that elute under acidic conditions.

During use, the buffer is applied to the affinity column after loading and washing to recover bound AAV. Eluate fractions are typically collected into, or promptly adjusted with, a suitable neutralizing buffer to return the sample to near-physiological pH. The buffer is compatible with downstream processing steps, including buffer exchange, desalting, tangential flow filtration, and ion-exchange chromatography.

  • 0.1M Acetic Acid
  • 0.4M Sodium Chloride
  • Adjusted to pH 2.75
  • AAV Affinity Chromatography Elution: Low-pH elution of AAV capsids from affinity resins (e.g., AVB/AAVX) during the capture step.

  • Non-Specific Binding Disruption: Acidic conditions reduce weak ionic and hydrophobic interactions to improve purity during elution.

  • Serotype-Compatible Elution: Effective across common AAV serotypes to recover intact capsids from affinity matrices.

  • Resin Stripping/Regeneration: Used post-run to strip tightly bound impurities from AAV affinity resins.

  • Process Development and Scale-Up: Standardized elution conditions for optimizing recovery, purity, and capsid integrity.

  • Fraction Collection for Downstream Buffer Exchange: Collection of eluted AAV fractions for immediate neutralization and buffer exchange into formulation buffers.

  • pH: 2.75 ± 0.10
