AAV Affinity CIP2 Solution, Non-Sterile is a cleaning-in-place reagent intended for affinity chromatography systems used in adeno-associated virus purification workflows. It is supplied non-sterile for routine column maintenance and turnover between purification runs, and is used to disrupt and remove residual biomaterials from affinity media and associated hardware surfaces.

The solution is applied as part of a defined CIP sequence that includes a pre-rinse, controlled exposure, and post-rinse with water or buffer as specified by the chromatography protocol. It is compatible with typical column materials used in bioprocessing and is intended for laboratory and manufacturing environments that follow established chemical handling and waste disposal procedures.

  • 6M Guanidine Hydrochloride
  • AAV Affinity Column Cleaning-in-Place (CIP): High-strength 6 M guanidine hydrochloride removes tightly bound impurities and fouling from AAV affinity resins between cycles.

  • Resin Regeneration: Restores capacity by denaturing and clearing host cell proteins, nucleic acids, and lipids from the resin matrix.

  • Aggregate and Precipitate Removal: Solubilizes AAV aggregates and protein precipitates on resin, frits, and hardware surfaces.

  • Endotoxin and Bioburden Reduction: Reduces endotoxin load and microbial bioburden (e.g., Escherichia coli, Pseudomonas aeruginosa); not a sterilant.

  • Sanitization of Downstream Hardware: Flush for compatible pumps, tubing, and valves to remove proteinaceous soils and carryover.

  • Carryover Mitigation: Minimizes cross-contamination between purification campaigns and product lots.

  • Cleaning Validation Studies: Used as a worst-case chaotropic agent to challenge and verify protein removal in CIP protocols.

  • Nucleic Acid Decontamination: Disrupts DNA/RNA interactions to clear residual host DNA and plasmid contaminants from columns.
