AAV Affinity Wash Buffer is a Tris-based, high-ionic-strength buffer formulated for wash steps during affinity purification of adeno-associated virus (AAV) particles.

The composition provides stable buffering and consistent salt conditions during column washing, supporting the removal of unbound and weakly associated impurities and compatibility with downstream processing.

  • 20mM Tris Base
  • 1.0M Sodium Chloride
  • Adjusted to pH 7.5
  • AAV Affinity Chromatography Wash: High-salt wash during capture to remove nonspecific proteins and nucleic acids while keeping AAV bound to the resin.

  • Column Equilibration and Conditioning: Used to equilibrate and condition AAV affinity media prior to sample loading for consistent binding performance.

  • Post-Load Cleansing Wash: Applied after sample loading to clear weakly bound host cell proteins and DNA without eluting AAV.

  • Sample Dilution/Adjustment: Mixed with lysate or supernatant to set ionic strength and pH before affinity capture.

  • Resin Specificity Enhancement: Minimizes nonspecific interactions on AVB/AAVX and similar AAV affinity resins to improve purity.

  • Serotype-Agile Processing: Compatible wash for multiple AAV serotypes during affinity capture workflows.

  • Carryover Reduction Between Runs: Used between cycles to limit residual contaminants and improve run-to-run consistency.

  • Endotoxin and Bioburden Mitigation: Helps reduce carryover of endotoxin and microbial impurities originating from E. coli-derived materials during wash steps.
