AAV Affinity Elution Buffer is an acidic, salt-containing buffer formulated for the elution of adeno-associated virus (AAV) particles from affinity chromatography media. The low pH promotes dissociation of capsid–ligand interactions, while ionic strength supports capsid stability during brief elution steps.

Following elution, fractions are typically collected on ice and promptly neutralized to a near-physiological pH, with subsequent buffer exchange as required for downstream applications.

  • 0.1M Acetic Acid
  • 0.4M Sodium Chloride
  • Adjusted to pH 2.75
  • AAV Affinity Chromatography Elution: Elutes recombinant AAV from affinity resins (e.g., AVB, AAVX) using low-pH 0.1M acetic acid/0.4M sodium chloride, pH 2.75.

  • Impurity Clearance During Elution: Disrupts nonspecific interactions to reduce host-cell protein and DNA carryover at the affinity step.
