AAV Filtration Buffer is a defined aqueous buffer designed for preparation, clarification, and filtration of adeno-associated virus samples. A Tris-based buffering system maintains stable pH during processing, sodium chloride provides appropriate ionic strength, sucrose supports osmotic stability of viral capsids, and Poloxamer 188 limits non-specific adsorption to filtration membranes and processing surfaces.

This buffer is suitable for workflows including tangential flow filtration, centrifugal and syringe filtration, buffer exchange, and sterile filtration steps, and is compatible with downstream concentration and storage of AAV preparations.

  • 20mM Tris Base
  • 0.2M Sodium Chloride
  • 1.0% Sucrose
  • 0.1% Poloxamer 188
  • Adjusted to pH 8.0
  • AAV Clarification and Sterile Filtration: Used as a process buffer to clarify and sterile-filter AAV preparations while maintaining capsid integrity.

  • Tangential Flow Filtration (TFF)/Diafiltration: Serves as the exchange buffer during AAV concentration and diafiltration for consistent ionic strength and pH control.

  • Formulation and Dilution of AAV: Provides a final formulation/dilution matrix (20 mM Tris, 0.2 M NaCl, 1.0% sucrose, 0.1% Poloxamer 188, pH 8.0) to stabilize vectors for downstream use.

  • Capsid Stabilization During Processing: Sucrose (1.0%) helps protect AAV capsids from shear and osmotic stress throughout filtration steps.

  • Reduced Adsorption and Improved Recovery: Poloxamer 188 (0.1%) minimizes AAV loss to tubing, filters, and vessel surfaces to enhance yield.

  • Chromatography Support: Used as an equilibration/wash buffer around pH 8.0 with moderate salt (0.2 M NaCl) during ion-exchange capture or polishing of AAV.

  • Sample Preparation for Analytical Assays: Maintains native conditions for qPCR/ddPCR titering, ELISA, and infectivity assays following filtration steps.

  • Membrane Conditioning and Wetting: Conditions virus-grade filters prior to load to reduce non-specific binding and improve throughput.

  • Intermediate Hold Buffer: Stabilizes AAV during short-term holds between unit operations at controlled temperature.

  • Process-Compatible Diluent: Used to adjust conductivity and pH of in-process pools without introducing denaturing components.
