YPD Agar Plates are a nutrient-rich solid medium for the cultivation, maintenance, and enumeration of yeasts and fungi. The formulation provides amino acids, peptides, vitamins, and a fermentable carbon source that support robust growth of organisms such as Saccharomyces cerevisiae and related species.

Plates are suitable for streaking, spread plating, and colony isolation in academic and industrial workflows. Incubate at organism-appropriate temperatures (commonly 25–30°C for many yeasts). No antibiotics or selective agents are included.

  • 1.0% Yeast extract
  • 2.0% Peptone
  • 2.0% Glucose
  • 1.5% Agar
  • Yeast Cultivation and Colony Isolation: Rich, non-selective medium for robust growth and single-colony isolation of Saccharomyces and other fungi.

  • Strain Maintenance and Stocking: Routine streaking, passaging, and plate storage of laboratory yeast strains.

  • Transformation Recovery: Post-transformation outgrowth prior to selection on defined or drug-containing media.

  • Replica Plating and Genetic Screens: Initial growth for phenotypic, temperature-sensitive, and suppressor/synthetic-lethal screens.

  • Mating and Ploidy Workflows: Pre-growth and lawn formation for mating assays and subsequent ploidy analyses.

  • Drug/Stress Spot Assays: Comparative growth under antifungals or stressors for sensitivity profiling.

  • Viability and CFU Enumeration: Quantification of viable yeast by plate counts and dilution spotting.

  • Fermentation Phenotype Checks: Rapid assessment of growth and morphology on glucose-rich plates.

  • Quality Control of Yeast Cultures: Monitoring culture purity and consistency during strain construction.

  • Enzyme and Reporter Assays: Plate-based readouts (e.g., colorimetric/fluorescent reporters) after growth on rich medium.
