Tryptic Soy Agar (TSA) Plates are a general-purpose, nonselective solid medium for cultivation and maintenance of a broad range of nonfastidious bacteria. The formulation is based on pancreatic digest of casein (tryptone), papaic digest of soybean meal (soytone), sodium chloride, and agar.

Used for routine culture, microbial enumeration, and observation of colony morphology from clinical, food, water, and environmental samples, including organisms such as Staphylococcus aureus, Escherichia coli, Bacillus subtilis, and Pseudomonas aeruginosa. Typical plates are incubated aerobically at 20–37 °C; the medium has a final pH of 7.3 ± 0.2 at 25 °C.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • General Cultivation: Broad-purpose growth of nonfastidious aerobic and facultative bacteria (e.g., Escherichia coli, Staphylococcus aureus, Bacillus subtilis, Pseudomonas aeruginosa, Enterococcus faecalis).

  • Colony Isolation and Enumeration: Streaking for isolated colonies and counting CFUs from clinical, environmental, and industrial samples.

  • Environmental Monitoring: Surface swab recovery and settle plates for air monitoring in cleanrooms, labs, and production areas.

  • Quality Control (Bioburden/Microbial Limits): Routine QC testing in pharma, biotech, and medical device manufacturing using soybean–casein digest agar.

  • Food and Beverage Testing: Total aerobic plate counts and recovery of stressed organisms from food, dairy, and beverage samples.

  • Water and Utility Monitoring: Heterotrophic plate counts and general microbial assessments of process water and rinsates.

  • Culture Maintenance: Subculturing, colony purification, and stock preparation for strain propagation and storage.

  • Disinfectant/Sanitizer Efficacy Studies: Neutralization and recovery medium for surface validation and cleaning verification.

  • Antimicrobial Screening (Non-clinical): Growth controls, lawn culture preparation, and basic zone-of-inhibition assays.

  • Education and Training: Teaching basic microbiology techniques, aseptic practice, and colony morphology.
