10% Tween-20 Solution is an aqueous preparation of the nonionic surfactant polysorbate 20 for laboratory use in buffer preparation, wash solutions, and sample processing. It reduces surface tension and aids dispersion of hydrophobic components in biochemical and cell-based workflows.

Typical applications include use as a wash additive in ELISA and Western blotting, as a blocking or permeabilizing component in immunostaining protocols, and for gentle solubilization of membrane-associated proteins. It is compatible with common buffer systems such as PBS, Tris, and saline, and is suitable for procedures involving organisms like E. coli where nonionic surfactants are required.

  • 10% (w/v) Tween-20

  • Western Blotting (TBST): Non-ionic surfactant added to wash buffers to reduce nonspecific antibody binding and background.

  • ELISA: Used in wash and blocking buffers to minimize nonspecific interactions and improve assay signal-to-noise.

  • Immunohistochemistry/Immunofluorescence: Included in wash/block buffers for gentle membrane permeabilization and background reduction.

  • Flow Cytometry: Added to wash buffers to limit cell clumping and decrease nonspecific antibody binding.

  • Protein Purification/Storage: Prevents protein aggregation and adsorption to plastics; stabilizes proteins in chromatography and storage buffers.

  • Immunoprecipitation/ChIP: Present in wash buffers to reduce nonspecific protein–protein and protein–DNA interactions.

  • PCR/qPCR Setup: Small amounts reduce surface adsorption of enzymes and templates, improving reaction consistency.

  • Cell Lysis/Permeabilization: Gentle detergent for extracting membrane proteins or permeabilizing cells without extensive denaturation.

  • Plate-Based Assays: Lowers surface tension to improve reagent spreading, reduce edge effects, and enhance wash efficiency.

  • General Buffer Formulation: Incorporated at working dilutions from a 10% (w/v) stock to control nonspecific binding and improve solubilization.
