LB Lennox Broth is a nutrient medium used for the cultivation and maintenance of laboratory strains of E. coli and other enteric bacteria. It is suitable for routine propagation, cloning workflows, and general microbiological applications in liquid culture.

The Lennox formulation employs a reduced sodium chloride level compared to LB Miller, supporting work where lower ionic strength is preferred. Antibiotics, inducers, or supplements may be added as required for plasmid maintenance or selection.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 0.5% Sodium Chloride
  • Routine Bacterial Cultivation: General growth and propagation of lab strains of Escherichia coli and other nonfastidious bacteria.

  • Plasmid Cloning and Amplification: Antibiotic-selected liquid cultures for plasmid maintenance and preparation (mini-/midi-/maxi-preps) in Escherichia coli.

  • Starter Cultures for Protein Expression: Overnight and seed cultures of Escherichia coli prior to induction in richer media.

  • Preparation of LB Agar Plates: Base medium (when supplemented with agar) for colony isolation, screening, and antibiotic selection.

  • Transformation Outgrowth: Short recovery of chemically competent or electrocompetent Escherichia coli after DNA uptake.

  • Bacteriophage Work: Host cultivation and phage propagation/titering (often with added Mg2+/Ca2+) in Escherichia coli systems.

  • Strain Maintenance and Archiving: Overnight cultures for preparing glycerol stocks and routine passaging of Escherichia coli.

  • Conjugation and Mating Assays: Donor/recipient culture growth for plasmid transfer experiments in Escherichia coli and related bacteria.
