2XYT (Yeast extract Tryptone) Agar in a bottle is a solid microbiological medium for the cultivation and maintenance of laboratory strains of E. coli and other compatible bacteria. It is based on the 2XYT formulation solidified with agar for plate-based applications.

Supplied as a sterile, meltable bottle for preparing agar plates or overlays. Suitable for routine propagation, colony isolation, and maintenance of E. coli on solid medium.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.50% Sodium Chloride
  • 1.5% Agar
  • Bacterial Plating and Colony Isolation: Solid medium for streaking and isolating single colonies of E. coli and other nonpathogenic lab strains.

  • Transformation Selection: Supports antibiotic selection of transformed E. coli colonies carrying recombinant plasmids.

  • Blue-White Screening: Used with X-gal/IPTG for lacZ α-complementation screening of cloning inserts in E. coli.

  • Phage Plaque Assays: Suitable agar base for M13 and filamentous phage titration and plaque formation on E. coli lawns.

  • Plasmid Maintenance: Growth and maintenance of recombinant plasmid-bearing E. coli strains on selective plates.

  • Colony PCR Sampling: Source plates for picking colonies as DNA templates for downstream PCR verification.

  • CFU Enumeration: Quantification of viable bacterial counts from liquid cultures or dilutions.

  • Strain Preservation and Tracking: Short-term storage and routine passaging of E. coli laboratory strains on agar plates.

  • Library Screening: Plating of combinatorial or phage display libraries for clone selection in E. coli.

  • General Microbiology Teaching and QC: Convenient ready-to-use agar for instructional labs and basic quality control checks.
