YPD Agar Plates with 2.5% Agar are a nutrient-rich agar medium formulated for the cultivation, isolation, and maintenance of Saccharomyces cerevisiae and other yeast species. Peptone and yeast extract provide peptides, amino acids, vitamins, and trace nutrients, while glucose serves as a readily metabolizable carbon source. The elevated agar content produces a firm surface suitable for streaking, colony isolation, and routine culture handling.

This medium is supplied without antibiotics or selective agents and is intended for general-purpose yeast work, including propagation, plating, and enumeration.

  • 1.0% Yeast Extract
  • 2.0% Peptone
  • 2.5% Agar
  • 2.0% Glucose
  • Yeast Culture and Colony Isolation: Nonselective growth and single-colony purification of Saccharomyces cerevisiae and other yeasts on rich medium (1.0% Yeast Extract, 2.0% Peptone, 2.0% Glucose, 2.5% Agar).

  • CFU Enumeration and Viability Testing: Quantification of viable yeast cells via colony counts.

  • Transformation Recovery and Outgrowth: Post-transformation recovery prior to selection or replica plating.

  • Growth Phenotyping and Spot Assays: Comparative fitness, temperature sensitivity, and drug-response spot tests.

  • Antibiotic/Drug Selection Base: Supplementation with G418, nourseothricin, or hygromycin B for selectable marker screening in yeast.

  • Mating and Patching: Mating assays, diploid selection steps, and routine patching or streaking for strain maintenance.

  • Replica Plating and Library Handling: Transferring colonies to selective media and managing mutant or deletion libraries.

  • Fermentation Strain Screening: Rapid plate-based evaluation of production strains and adaptive evolution lineages.

  • Contamination Assessment: Visual detection of microbial contaminants in yeast cultures, including potential growth of Escherichia coli if present.

  • Teaching and Training: General microbiology and yeast genetics exercises and plate technique practice.
