YPD Agar Plates are a complex, non-selective medium used for the cultivation and plating of yeasts such as Saccharomyces cerevisiae and related species. The formulation includes yeast extract and peptone as sources of vitamins, amino acids, peptides, and other growth factors, with glucose (dextrose) as the primary carbon source. Agar provides a solid surface for colony isolation and routine maintenance.

This medium is commonly employed for general propagation, viability assessment, and genetic manipulations where a rich, undefined nutrient base is required for robust yeast growth.

  • 1.0% Yeast Extract
  • 2.0% Peptone
  • 2.0% Cerelose Anhydrous Dextrose (Glucose)
  • 2.0% Agar
  • Yeast Culture and Colony Isolation: Routine streaking and single-colony isolation of Saccharomyces cerevisiae and other yeasts on rich medium (1.0% yeast extract, 2.0% peptone, 2.0% dextrose, 2.0% agar).

  • Yeast Strain Maintenance: Short- and medium-term storage of yeast stocks as patches or slants for subsequent experiments.

  • Post-Transformation Recovery and Plating: Outgrowth and initial plating of yeast transformants prior to transfer to selective media.

  • Antibiotic Selection in Yeast: Plating with antibiotics (e.g., G418, hygromycin B, nourseothricin) for selecting engineered yeast carrying resistance markers.
