YPAD Broth is a rich yeast medium formulated for cultivation of Saccharomyces cerevisiae and related yeasts. The combination of peptone, yeast extract, and glucose provides amino acids, vitamins, nitrogen, and a readily metabolizable carbon source for routine propagation and biomass production.

The inclusion of adenine hemisulfate supplements purine biosynthesis and minimizes red pigment accumulation in ade mutants, supporting growth of adenine-requiring strains. Suitable for culture expansion, strain maintenance, and recovery after genetic manipulation.

  • 2.0% Peptone
  • 1.0% Yeast Extract

  • Yeast Culturing and Propagation: Rich, non-selective medium for robust growth of Saccharomyces cerevisiae and other yeasts.

  • Starter Cultures for Protein Expression: Generates high-density inocula for downstream expression in yeast systems.

  • Transformation Recovery/Outgrowth: Supports rapid post–LiAc/PEG or electroporation recovery prior to plating on selective media.

  • Colony Formation and Replica Plating: Promotes fast colony development and reliable transfer to selective/dropout plates.

  • Ade Mutant Phenotype Maintenance: 0.006% adenine hemisulfate suppresses red pigmentation in ade1/ade2 strains for clearer phenotype assessment.

  • Growth Curves and Physiology: Enables reproducible growth-rate measurements and nutrient response studies using 2.0% peptone, 1.0% yeast extract, 2.0% glucose.

  • Antimicrobial/Drug Sensitivity Assays: Serves as a rich baseline medium for testing compound effects on yeast viability.

  • Pre-cultures for Genetic Screens: Used to expand strains prior to selection on defined dropout media or assay plates.
