Yeast Nitrogen Base (YNB) Agar is a defined agar medium for cultivation, maintenance, and physiological characterization of yeasts in the presence of a single carbon source and an inorganic nitrogen source. The formulation supplies vitamins, minerals, and trace elements, with glucose as the carbon source and ammonium sulfate as the nitrogen source. The agar matrix enables colony isolation, replica plating, and assessments of carbon and nitrogen utilization.

These ready-to-use plates are suitable for growth of laboratory yeast strains such as Saccharomyces cerevisiae and for selection experiments when supplemented with required amino acids, bases, or other defined components as needed for specific auxotrophies.

  • 2.0% Cerelose Anhydrous Dextrose (Glucose) 0.5% Ammonium Sulfate
  • Yeast Strain Maintenance: Defined minimal agar for routine cultivation of Saccharomyces and Candida using 2.0% glucose and 0.5% ammonium sulfate.

  • Auxotrophic Selection: Selection of transformants (e.g., URA3, LEU2, HIS3, TRP1 markers) when used with appropriate dropout supplements.

  • Plasmid Maintenance: Stable selection and propagation of plasmid-bearing yeast under defined nutritional conditions.

  • Colony Isolation and Enumeration: Streaking, single-colony isolation, and CFU counts on a standardized minimal medium.

  • Yeast Two-Hybrid Screening: Growth and reporter selection on SD-based selective plates (e.g., -Leu/-Trp/-His) prepared with this YNB agar base.

  • Metabolic and Carbon Utilization Studies: Baseline growth on 2.0% glucose for comparative metabolic assays and nutrient limitation studies.

  • Phenotypic Screening: Assessment of growth under stressors (e.g., pH, osmotic, drug exposure) in a chemically defined matrix.

  • Quality Control of Yeast Cultures: Purity checks and verification of strain identity on minimal, reproducible agar.
