2XYT Broth is a nutrient-rich yeast extract–tryptone medium for cultivation of E. coli and related bacteria. The balanced composition provides peptides, amino acids, vitamins, and minerals suitable for aerobic shaking cultures and routine propagation.

Common applications include growth of recombinant E. coli for plasmid maintenance, cloning workflows, and protein expression, with optional supplementation (e.g., antibiotics) according to experimental requirements.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • Plasmid DNA Production: High-yield cultivation of E. coli for mini/midi/maxi-prep plasmid workflows.

  • Recombinant Protein Expression: Supports robust IPTG-induced expression in E. coli shake flasks and bioreactors.

  • Phage Propagation: Amplification of M13 and other filamentous phage using E. coli host strains.

  • Antibiotic Selection and Cloning: Growth and selection of transformed E. coli carrying antibiotic-resistant plasmids.

  • Transformation Recovery: Post-heat shock or electroporation outgrowth for E. coli to express antibiotic resistance markers.

  • Starter and Overnight Cultures: Preparation of dense E. coli inocula for downstream fermentation or expression runs.

  • Library Amplification: Expansion of plasmid, cDNA, and phage display libraries in E. coli.

  • Glycerol Stock Preparation: Generating stable freezer stocks of laboratory E. coli strains.

  • Nucleic Acid Preparation: Growth of E. coli to produce biomass for high-yield DNA/RNA extraction.

  • General Bacterial Cultivation: Routine propagation and maintenance of cloning and protein-production E. coli strains.
