2XYT (Yeast extract Tryptone) Broth with 0.1% Glucose is a nutrient-rich liquid medium used for cultivation of laboratory strains of E. coli and related bacteria. The 0.1% glucose supplies a readily utilizable carbon source during early culture growth.

Suitable for preparation of liquid cultures and for addition of selective agents as required by the experimental design. Not for clinical or diagnostic use.

  • 1.6% Tryptone
  • 1.0% Yeast Extract
  • 0.5% Sodium Chloride
  • 0.1% Cerelose Anhydrous Dextrose (Glucose)
  • High-Density Cultivation of Escherichia coli: Rich 2xYT formulation supports rapid, robust growth for routine culture and scale-up.

  • Plasmid DNA Preparation (Mini/Midi/Maxi): Produces high-yield overnight cultures for plasmid propagation with improved plasmid stability.

  • Recombinant Protein Expression (T7/lac Systems): 0.1% glucose suppresses basal expression prior to induction, enhancing expression control.

  • Phage Work (M13/Filamentous Phage): Common medium for phage propagation, infection, and titering using E. coli hosts.

  • Antibiotic Selection and Plasmid Maintenance: Supports reliable selection and retention of plasmids during growth.

  • Post-Transformation Recovery: Used for outgrowth of chemically competent or electrocompetent E. coli to maximize colony formation.

  • Starter Cultures and Seed Inocula: Ideal for generating inoculums for larger shaker, bioreactor, or fermenter runs.

  • General Cloning Workflows: Supports cloning, subcloning, and library amplification with reduced leaky lac activity due to glucose.
