X-Gal is a chromogenic substrate for β-galactosidase used to visualize lacZ activity. In cloning workflows, it enables blue-white screening of recombinant colonies via α-complementation of lacZ on agar plates supplemented with IPTG and a selective antibiotic, allowing visual differentiation of insert-containing transformants in E. coli.

Typical working concentrations range from 20–40 µg/mL in agar media. Prepare solutions in an appropriate solvent, protect from light, and store at low temperature to maintain stability. Applications include colony screening, reporter assays, and detection of β-galactosidase activity in E. coli and related systems.
