amdS Biolistic Agar Plates are a defined minimal medium formulated for the selection of amdS-positive transformants following particle bombardment. The medium uses acetamide as the sole nitrogen source to enable selection based on acetamidase activity, with glucose as the primary carbon source and Noble Agar as the solidifying agent.

The formulation is buffered with phosphate and supplemented with cesium chloride, calcium and magnesium salts, and trace metals including iron, manganese, zinc, and cobalt to support growth and selection on minimal conditions.

  • 2.00% Noble Agar
  • 2.00% Glucose
  • 1.50% Monopotassium Phosphate
  • 0.17% Cesium Chloride
  • 0.06% Acetamide
  • 0.06% Calcium Chloride
  • 0.06% Magnesium Sulfate
  • 5.0µg/mL Ferrous Sulfate
  • 1.6µg/mL Manganese Sulfate
  • 1.4µg/mL Zinc Sulfate
  • 1.0µg/mL Cobalt Chloride

  • Biolistic Transformation Recovery: Used as recovery and plating medium after particle bombardment of filamentous fungi carrying the amdS selectable marker.

  • Positive Selection of amdS Transformants: Acetamide-based selection to isolate amdS+ clones in Aspergillus and related filamentous fungi.

  • CRISPR/Cas9 Genome Editing Workflows: Selection of edited strains when amdS is used as the selectable marker.

  • Gene Knockout/Knock-in Screening: Identification and cleanup (re-streaking) of targeted transformants under amdS selection.

  • Strain Stabilization and Maintenance: Routine propagation of amdS-marked fungal strains under defined, selective conditions.

  • Single-Spore Isolation of Transformants: Purification of individual amdS+ colonies from mixed populations following transformation.

  • Plasmid Integration Verification: Growth-based confirmation of selectable marker presence prior to molecular validation.

  • Phenotypic Assays on Defined Medium: Assessment of growth and morphology on a minimal, trace-element–supplemented, glucose/acetamide medium.
