8M Urea Solution, Non-Sterile is a chaotropic reagent used for protein denaturation, nucleic acid purification, and disruption of noncovalent interactions. Typical applications include solubilization of proteins from inclusion bodies in E. coli, preparation of denaturing electrophoresis buffers, and RNase inactivation workflows.

Supplied non-sterile; filter-sterilize if sterility is required. Store at 2–8 °C and minimize exposure to elevated temperature or alkaline pH to limit cyanate formation; avoid repeated freeze–thaw cycles.

  • 8M Urea Solution

  • Denaturing PAGE (Urea-PAGE): 8M Urea Solution is used to denature RNA and ssDNA for high-resolution polyacrylamide gel electrophoresis.

  • Nucleic Acid Hybridization: Adjusts stringency in Northern, Southern, and in situ hybridization workflows.

  • DNA Sequencing Gels: Provides denaturing conditions for Sanger polyacrylamide sequencing gels.

  • Protein Denaturation/Solubilization: Acts as a chaotrope to unfold and solubilize proteins for analysis or refolding screens.

  • 2D Gel Electrophoresis Prep: Used in sample buffers (urea/thiourea) to solubilize proteins prior to IEF and SDS-PAGE.

  • Inclusion Body Extraction: Facilitates chaotropic solubilization of aggregates from Escherichia coli expression systems.

  • RNase/Nuclease Inactivation: Denatures nucleases to help preserve RNA integrity during preparation.

  • Nucleic Acid Purification Buffers: Included in denaturing buffers for cleanup of RNA and ssDNA.

  • Probe and Oligo Preparation: Maintains single-stranded states for labeling, annealing control, and melting studies.

  • Membrane Protein Solubilization: Aids in disrupting hydrophobic interactions for extraction and analysis.
