5.6 mg/mL Uracil Supplement is a ready-to-use uracil stock for defined media and culture workflows. It supplies exogenous uracil to support the propagation of uracil-auxotrophic strains and to terminate uracil-based selection when required.

Applications include supplementing minimal or dropout media, maintaining strains that require uracil for growth, and workflows involving URA3 selection or 5-FOA counter-selection in yeast. Compatible with uracil-dependent strains of E. coli and Saccharomyces cerevisiae.

  • 5.6mg/mL Uracil Solution

  • Auxotrophic Strain Cultivation: Supplements minimal media to support growth of uracil auxotrophs (e.g., Saccharomyces cerevisiae ura3Δ, Schizosaccharomyces pombe ura4Δ, Candida albicans ura3Δ, Escherichia coli ΔpyrF).

  • Yeast Genetic Selection: Preparation of SD+Ura media for complementation tests, plasmid maintenance/loss assays, and replica plating.

  • 5-FOA Counterselection: Added with 5-fluoroorotic acid to select against URA markers while supporting growth of Ura− cells.

  • Transformation Workflows: Ensures recovery and propagation of uracil-requiring transformants and URA-marker plasmids.

  • CRISPR/Cas Editing: Used with URA3/ura3 or pyrF/pyrF− selectable and counterselectable marker schemes.

  • Minimal Media Supplementation: Standard additive for defined media in microbial culture and fermentation studies, including E. coli auxotrophs.

  • Nucleotide Metabolism Assays: Provides defined uracil levels for salvage-pathway and pyrimidine biosynthesis studies.
