1X TBS, Modified is an isotonic, non-denaturing Tris-buffered saline suitable for routine laboratory procedures requiring a stable ionic environment. It is commonly used for membrane and slide washing, tissue and cell rinsing, and preparation of protein and antibody solutions.

This buffer serves as a base for TBS-T when supplemented with a nonionic detergent and is compatible with workflows in western blotting, ELISA, and immunohistochemistry.

  • 50mM Tris-HCl
  • 150mM Sodium Chloride
  • Western Blotting: Antibody dilution and membrane washing; add Tween-20 (TBST) to reduce nonspecific binding.

  • ELISA: Plate washing and antibody/sample dilution to maintain isotonicity and stable pH.

  • Immunohistochemistry/Immunocytochemistry: Tissue/cell washing and blocking buffer base to minimize background.

  • Immunofluorescence Microscopy: Wash buffer for slides and coverslips to preserve antigen–antibody interactions.

  • Immunoprecipitation/Co-IP: Wash buffer to reduce nonspecific protein interactions during capture.

  • Protein Biochemistry: Sample dilution, reagent preparation, and column equilibration/wash when phosphate is undesirable.

  • Cell and Tissue Handling: Isotonic rinse for cells and tissues prior to fixation or lysis.

  • General Laboratory Buffer: Stable pH and ionic strength (50 mM Tris-HCl, 150 mM NaCl) for compatible enzyme assays and reagent formulation.
