Tris–magnesium chloride–sucrose buffer at pH 8.0 for molecular biology workflows. Provides buffering capacity at alkaline pH, supplies Mg2+ for DNA- and RNA-associated enzymes, and includes sucrose for osmotic stabilization.

Applications include preparation and handling of competent E. coli cells, maintenance of bacteriophage stocks, dilution or storage of nucleic acids and protein–nucleic acid complexes, and general-purpose reaction setup where divalent cations and osmotic support are required.

  • PCR and qPCR: Provides pH 8.0 buffering and 2 mM Mg2+ during reaction setup or dilution of Mg-free components.

  • Restriction Digests and DNA Processing: Serves as a dilution/storage buffer for DNA and enzymes that require Mg2+ prior to adding specific reaction buffers.

  • Enzyme Assays: Supports Mg2+-dependent enzymes (e.g., polymerases, nucleases, kinases) with stable pH and osmoprotection from 4% sucrose.

  • Cell and Protoplast Handling: Used as an osmoprotective resuspension/wash buffer for bacteria such as E. coli and Bacillus subtilis, helping stabilize membranes during centrifugation and manipulation.

  • Bacteriophage Work: Employed for dilution or short-term storage of phage preparations where Mg2+ and sucrose help maintain particle stability.

  • Protein Stabilization: Used as a storage/dilution buffer for Mg2+-binding proteins and complexes, with sucrose aiding stability during handling and freeze-thaw.
