Tryptic Soy Agar (TSA) is a general-purpose agar medium formulated for the cultivation and maintenance of nonfastidious microorganisms. Enzymatic digests of casein and soybean provide peptides, amino acids, and growth factors, sodium chloride maintains osmotic balance, and agar supplies a firm surface suitable for colony isolation and morphological observation.

TSA is commonly used for routine cultivation and enumeration of bacteria in laboratory settings, including organisms such as Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa. Plates are typically prepared by sterilizing the hydrated medium by autoclaving and pouring molten agar into sterile petri dishes for subsequent inoculation and incubation.

  • General-Purpose Cultivation: Supports robust growth of a wide range of nonfastidious bacteria (e.g., Escherichia coli, Staphylococcus aureus) for routine culture and maintenance.

  • Environmental Monitoring: Used for surface and air (settle plate) monitoring in labs, food production, and cleanrooms to detect aerobic microbial contamination.

  • Microbial Enumeration: Suitable for total aerobic plate counts from water, foods, and cosmetic/pharmaceutical samples.

  • Colony Isolation and Morphology: Enables streak plating and observation of colony characteristics for organisms such as Pseudomonas aeruginosa and Bacillus subtilis.

  • Quality Control Strain Maintenance: Routine propagation and storage of reference strains (e.g., Staphylococcus aureus ATCC, Escherichia coli ATCC) for QC workflows.

  • Sanitation and Disinfectant Efficacy Testing: Recovery of surviving organisms after cleaning validations to assess bioburden reduction.

  • Food and Beverage Testing: Recovery and enumeration of aerobic bacteria including Salmonella enterica (with appropriate enrichment steps) and spoilage organisms.

  • Pharmaceutical and Cosmetic Testing: Bioburden assessment and growth promotion testing to verify medium performance.

  • Water and Environmental Samples: Cultivation and count of heterotrophic bacteria from potable and process water.

  • Antimicrobial Screening (Non-standard): Preliminary growth-based checks or disk diffusion setup when Mueller–Hinton is not required, including testing against Listeria monocytogenes and Pseudomonas aeruginosa for recovery.
