Tryptic Soy Agar is a general-purpose agar medium for cultivation and maintenance of nonfastidious microorganisms. It contains enzymatic digests of casein and soybean meal as sources of amino acids and peptides, sodium chloride to maintain osmotic balance, and agar as a solidifying agent.

Typical uses include routine cultivation, colony isolation, and plate counts of organisms such as Escherichia coli, Staphylococcus aureus, Bacillus subtilis, and Pseudomonas aeruginosa.

  • 1.5% Tryptone
  • 0.5% Soytone
  • 0.5% Sodium chloride
  • 1.5% Agar
  • General Cultivation and Isolation: Broad-purpose growth medium for nonfastidious microbes, supporting organisms such as Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa.

  • Colony Isolation and Morphology: Streaking to obtain pure cultures and assess colony characteristics of bacteria like Bacillus subtilis and Enterococcus faecalis.

  • Enumeration (CFU Counts): Spread/pour plate methods for microbial load testing in water, food, and environmental samples (e.g., Salmonella enterica, Listeria monocytogenes).

  • Environmental Monitoring: Settle plates and contact plates for air and surface bioburden in cleanrooms and production areas.

  • Maintenance and Subculturing: Routine propagation and short-term storage of laboratory strains on plates or slants.

  • Quality Control (Growth Promotion Testing): Verification of medium performance using reference strains like Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa.

  • Food, Beverage, and Cosmetic Testing: Bioburden assessment and recovery of stressed or injured bacteria after processing.

  • Clinical and Diagnostic Microbiology: Primary culture of nonfastidious bacteria from specimens prior to identification workflows.

  • Blood Agar Base: Supplemented with blood to prepare hemolysis plates for enhanced recovery and differential observation.

  • Sterility and Membrane Filtration Recovery: Recovery medium for organisms captured by filters in pharmaceutical and water testing.
