SM Buffer is a buffered saline solution used for preparation, dilution, and storage of bacteriophages during work with E. coli hosts. The buffering system maintains near-neutral pH, monovalent and divalent ions support phage stability and adsorption, and a protein additive minimizes phage loss due to adherence to surfaces.

  • 100mM Sodium Chloride
  • 8mM Magnesium Sulfate
  • 0.01% Gelatin
  • 50mM Tris-HCl
  • Bacteriophage Storage and Stabilization: Maintains phage integrity during short- and long-term storage, leveraging Tris-HCl buffer, Mg2+ for capsid stability, and gelatin to reduce adsorption.

  • Phage Dilution and Transport: Used as an isotonic diluent for accurate serial dilutions and safe transport of phage samples.

  • Plaque Assays and Titer Determination: Employed as a diluent and recovery buffer for phage plating on Escherichia coli lawns.

  • Elution from Agar Plugs/Plates: Facilitates efficient recovery of phage from top agar plugs, plaques, and environmental samples.
