SC Minimal Glucose Agar Plates minus Uracil are defined yeast selection plates that support the growth and selection of uracil-prototrophic Saccharomyces cerevisiae strains or transformants carrying URA3. The medium contains 2% glucose as the carbon source and a complete supplement mixture lacking uracil, solidified with agar; omission of uracil selects for functional URA3 to maintain URA3-marked plasmids or chromosomal integrations.

  • Carbon: 2.0% Glucose
  • Nitrogen: 0.5% Ammonium Sulfate
  • Minus: Uracil and Glutamic Acid
  • Auxotrophic Selection (-Ura): Selects and maintains yeast strains carrying URA3-marked plasmids or integrations on uracil-dropout media.

  • Plasmid Maintenance: Ensures stable retention of URA3-based vectors in Saccharomyces cerevisiae under selective pressure.

  • Yeast Transformation Screening: Enables colony isolation and verification of URA3-positive transformants after DNA introduction.
