SOB Medium is a nutrient-rich broth used for cultivation and recovery of E. coli cells during cloning workflows, including post-transformation outgrowth prior to selection. Peptone and yeast-derived nutrients supply amino acids, peptides, and growth factors, while sodium and potassium salts maintain ionic strength and osmotic balance.

Magnesium chloride and magnesium sulfate provide divalent cations commonly included for transformation workflows with E. coli. SOB Medium is prepared as a clear, sterile solution intended for use as a liquid growth medium.

  • 2.0% Tryptone
  • 0.5% Yeast Extract
  • 10mM Sodium Chloride
  • 2.5mM Potassium Chloride
  • 10mM Magnesium Chloride
  • 10mM Magnesium Sulfate
  • Bacterial Transformation Recovery: Rich, magnesium-supplemented medium for rapid outgrowth of transformed E. coli prior to plating (pre-SOC step).

  • Electroporation and Heat-Shock Outgrowth: Supports recovery and expression of antibiotic resistance in newly transformed E. coli.

  • Preparation of Competent Cells: Used during workflows to generate high-efficiency competent E. coli cells.

  • Phage Propagation: High Mg2+ content supports infection and amplification of filamentous phage (e.g., M13) in E. coli.

  • Library Construction and Cloning: Facilitates recovery of high-complexity DNA libraries and high-efficiency cloning in E. coli.

  • Recombinant Strain Cultivation: Short-term growth of engineered E. coli prior to selection or screening.

  • Antibiotic Selection Readiness: Promotes rapid expression of resistance markers before applying selective pressure in E. coli.

  • Phage Display Workflows: Supports infection, panning, and amplification steps involving phage-infected E. coli.
