Sucrose Selection Agar Plates are a complex agar medium used for sacB-based counter-selection in Gram-negative bacteria such as E. coli. A Soytone and yeast extract nutrient base supports growth on a solid agar surface.

Sucrose provides selection against sacB-expressing cells, and L-(+)-arabinose enables induction of PBAD-controlled sacB for negative selection in workflows such as allelic exchange and plasmid curing.

  • 1.0% Soytone
  • 0.5% Yeast Extract
  • 1.5% Agar
  • 0.2% L-(+)-Arabinose
  • 6.0% Sucrose
  • sacB-Based Counterselection for Allelic Exchange: Selects for loss of sacB-containing integrations in Gram-negative hosts (e.g., Escherichia coli, Pseudomonas aeruginosa, Vibrio cholerae, Salmonella enterica) using 6.0% sucrose lethality.

  • pBAD-sacB Induction: 0.2% L-(+)-Arabinose induces sacB under pBAD control to enable negative selection during recombineering workflows.

  • CRISPR-Assisted Editing Negative Selection: Enriches for correctly edited clones by counterselecting sacB-marked intermediates after CRISPR/recombineering.

  • Plasmid Curing of sacB-Tagged Vectors: Eliminates sacB-bearing suicide or helper plasmids following integration/excision cycles.

  • Screening Post-λ Red or Flp-FRT Steps: Identifies double-crossover recombinants after marker exchange or cassette excision.

  • Transposon Mutagenesis Workflows: Resolves insertions when transposon delivery or helper constructs carry sacB for counterselection.

  • Isolation of Sucrose-Resistant Variants: Screens for sacB loss-of-function or recombination events that confer growth on 6.0% sucrose.
