Selective scFv Induction Broth SG/R minus Tryptophan, Uracil is a defined synthetic medium formulated with galactose and raffinose for cultivation and GAL-promoter–driven induction of single-chain variable fragment (scFv) expression in Saccharomyces cerevisiae. The omission of tryptophan and uracil enables selection of yeast strains or transformants complemented by TRP1 and URA3 markers.

This liquid medium supports growth and induction steps in yeast display and secretion workflows that require auxotrophic selection and galactose-based induction while minimizing glucose-mediated repression.

  • Selective scFv Induction Broth SG/R minus Tryptophan and Uracil
  • Galactose-Induced scFv Expression in Yeast: Drives scFv production from GAL promoters in Saccharomyces cerevisiae using Selective scFv Induction Broth SG/R minus Tryptophan and Uracil.

  • Dual Plasmid Selection (TRP1, URA3): Maintains selection for TRP1- and URA3-marked plasmids during induction by omitting tryptophan and uracil.

  • Yeast Surface Display Screening: Supports growth and induction for flow cytometry sorting (FACS) of scFv display libraries.

  • Affinity Maturation and Panning: Enables iterative binding selections of displayed scFv variants during galactose-driven expression.

  • Secreted scFv Production: Provides selective induction conditions for small-scale secretion of scFv for downstream assays.

  • Expression Optimization Studies: Used to optimize induction time, temperature, and media conditions for maximal scFv yield and functionality.

  • Maintenance of Selective Pressure During Induction: Preserves plasmid retention while inducing scFv in shake flasks or bioreactors.

  • Binding and Functional Assays Post-Induction: Produces selectively induced cultures for ELISA, flow binding, and activity assays of scFv.
