20X SSC, Molecular Biology Grade Solution is a concentrated saline-sodium citrate buffer used for nucleic acid blotting and hybridization workflows. When diluted to working strength, it provides defined ionic strength and buffering capacity for capillary transfer to membranes, prehybridization and hybridization steps, and controlled stringency washes on nylon or nitrocellulose substrates.

The solution is prepared with molecular biology grade reagents and filtered for clarity. It can be diluted to common working concentrations (for example, 10X, 5X, 2X, or 1X) according to protocol requirements for Southern, Northern, or in situ hybridization procedures.

  • 3M Sodium Chloride
  • 300mM Citric Acid
  • Trisodium Salt
  • Nucleic Acid Hybridization (Southern/Northern): Standard buffer for pre-hybridization, probe hybridization, and post-hybridization washes; commonly used at 1X–6X to control stringency.

  • Stringency Washes: Employed at defined dilutions (e.g., 2X, 1X, 0.5X, 0.2X, 0.1X) to modulate nucleic acid duplex stability during post-hybridization washing.

  • In Situ Hybridization (ISH/FISH): Used in tissue/cell hybridizations for probe dilution and washing steps to maintain ionic strength and pH.

  • Microarray Hybridization: Utilized in array pre-hybridization, probe hybridization, and wash buffers to support specific nucleic acid binding.

  • Blotting and Capillary Transfer: Provides ionic strength during transfer of DNA/RNA from gels to membranes in Southern and Northern blotting workflows.

  • Membrane Prehybridization/Blocking: Serves as a base buffer for blocking agents before probe addition to reduce nonspecific binding.

  • Nucleic Acid Resuspension/Storage: Used to dilute and store DNA/RNA to maintain stability and minimize aggregation.

  • Colony and Plaque Lifts: Applied during nucleic acid lifts from E. coli colonies or phage plaques onto membranes for subsequent hybridization.

  • RNase Protection/Northern Prep: Used as a wash and dilution buffer in RNA handling workflows requiring controlled ionic conditions.

  • Probe Preparation/Dilution: Provides a consistent ionic environment for diluting labeled DNA/RNA probes prior to hybridization.
