SB Media without Glucose is a rich, MOPS-buffered broth medium intended for the cultivation of E. coli in molecular biology and microbiology applications. This formulation contains no added glucose.

Common uses include routine growth of recombinant E. coli strains and propagation of plasmid-bearing cultures when supplemented with appropriate antibiotics as required.

  • 3.0% Tryptone
  • 2.0% Yeast Extract
  • 1.0% MOPS
  • High-Density Cultivation of Escherichia coli: Rich, MOPS-buffered medium (3.0% Tryptone, 2.0% Yeast Extract, 1.0% MOPS) supports rapid, high-cell-density growth for downstream workflows.

  • Plasmid DNA Production: Enables robust plasmid amplification in Escherichia coli for mini/midi/maxi-preps with reduced acidification due to MOPS buffering.

  • Recombinant Protein Expression in E. coli: Supports high-yield expression; absence of glucose minimizes catabolite repression for IPTG/lactose-based induction.

  • Seed Cultures for Fermentation: Used to generate dense, healthy starter cultures for bioreactors and scale-up.

  • Phage Work (M13/Phage Display): Supports propagation of filamentous phage in Escherichia coli host strains for display and amplification.

  • Antibiotic Selection and Cloning Workflows: Facilitates rapid growth of transformants under selective pressure for cloning and construct verification.

  • Overnight Cultures for Minipreps: Produces robust overnight growth of E. coli with stable pH for consistent plasmid yields.

  • Recovery and Outgrowth Post-Transformation: Rich, buffered environment promotes recovery of electroporated or chemically transformed E. coli.

  • Glycerol Stock Preparation: Generates high-density cultures suitable for long-term cryopreservation of E. coli strains.

  • Method Development and Media Optimization: Serves as a glucose-free, buffered baseline for testing carbon supplementation and induction strategies.
