Starch Agar Plates are a solid microbiological medium used for the cultivation of bacteria and the qualitative detection of extracellular amylase activity via starch hydrolysis. After incubation, flooding the plate with iodine produces a blue-black complex with intact starch, while clear zones around colonies indicate starch degradation. This medium is commonly applied in screening environmental or laboratory isolates, including amylase-positive species such as Bacillus subtilis and related Bacillus spp.

  • 1.0% Soluble Starch
  • 0.3% Beef Extract
  • 1.5% Agar
  • Amylase (Starch Hydrolysis) Test: Detects alpha-amylase production by flooding plates with iodine and observing clear zones around colonies.

  • Bacterial Differentiation: Differentiates amylolytic organisms such as Bacillus, Clostridium, Corynebacterium, Bacteroides, and Enterococcus based on starch degradation.

  • Industrial Strain Screening: Screens environmental and clinical isolates for high amylase production for biotechnology and enzyme manufacturing.

  • Quality Control of Enzyme Producers: Verifies amylase activity in production strains and validates enzyme lots or process consistency.
