1X RPMI-1640 Media without Glucose, Glycine, and Serine is a basal cell culture medium in which the carbohydrate source and the amino acids glycine and serine are intentionally omitted. This formulation supports experimental designs that require controlled nutrient supplementation, metabolic tracing, or selective omission of these components.

Use in workflows where glucose, glycine, and serine are supplied independently or where their absence is required. This medium is compatible with standard RPMI-1640 supplements such as sodium bicarbonate, L-glutamine or stable L-alanyl-L-glutamine, and antibiotics, according to application needs and cell line requirements.

  • 1X RPMI-1640 without Cerelose Anhydrous Dextrose (Glucose)
  • Glycine
  • Serine
  • Serine/Glycine Deprivation Studies: Supports assays probing one-carbon metabolism, nucleotide synthesis, and redox balance by omitting serine and glycine.

  • Glucose-Free Metabolism Experiments: Enables evaluation of glycolytic dependence, Warburg effect, and glycolysis inhibitor responses with controlled carbon source add-back (e.g., galactose).

  • Stable-Isotope Tracing (13C/15N): Facilitates clean labeling experiments with 13C-glucose, 13C-serine, or 13C-glycine for metabolic flux analysis.

  • Nutrient Add-Back and Rescue Screens: Allows systematic reintroduction of glucose, serine, glycine, formate, or folate to identify pathway dependencies.

  • Auxotrophy and Pathway Characterization: Used to assess serine/glycine auxotrophies and engineered metabolic pathways in mammalian cells.

  • Process Development for Hematopoietic Cells: Applied to T cells, hybridomas, and other hematopoietic lines under defined carbohydrate and amino acid conditions.

  • Drug Mechanism and Sensitivity Testing: Optimizes evaluation of antifolates and one-carbon pathway inhibitors (e.g., SHMT, MTHFD) under targeted nutrient limitations.

  • Radiolabeled Uptake/Incorporation Assays: Provides a defined background for tracing labeled precursors into nucleotides and lipids.

  • Genetic Screens Under Nutrient Stress: Enables CRISPR/RNAi screens with glucose or serine/glycine restriction to reveal metabolic liabilities.

  • Bioenergetics and Mitochondrial Function Tests: Supports assays (e.g., Seahorse) to dissect glycolysis versus oxidative phosphorylation with controlled substrate add-back.
