RPMI 1640 Medium without Glutamine and Glucose is a basal medium for in vitro culture of mammalian cells. The omission of Cerelose anhydrous dextrose (glucose) and L-glutamine allows defined supplementation and control of carbon and nitrogen sources.

This medium provides amino acids, vitamins, inorganic salts, and phenol red in a sodium bicarbonate–buffered system for use with CO2 incubation. Supplement with an appropriate carbohydrate (e.g., glucose) and a glutamine source (e.g., L-glutamine or stable dipeptide alternatives) prior to use, and sterilize by filtration after supplementation. It is suitable for the propagation and maintenance of human and animal lymphoid cell lines and primary cells under glucose- or glutamine-modulated conditions.
