20× PBS with 0.05% Tween‑20 is a concentrated phosphate‑buffered saline containing a nonionic surfactant for preparing working‑strength PBS‑T wash and rinse solutions used in immunoassays, blotting workflows, and general laboratory procedures.

Dilute 1:20 with reagent‑grade water to obtain 1× PBS‑T (final Tween‑20 at 0.0025%). Suitable for routine washing steps, sample handling, and buffer preparation where a phosphate buffer with low‑level nonionic surfactant is required.

  • 2.7M Sodium Chloride
  • 54mM Potassium Chloride
  • 28mM Potassium Phosphate Monobasic Anhydrous
  • 86mM Sodium Phosphate Dibasic Anhydrous
  • 0.05% Tween 20
  • Western Blotting: Washes membranes to remove nonspecific antibody binding during immunoblotting.

  • ELISA: Serves as a plate wash buffer to minimize background and enhance assay specificity.

  • Immunofluorescence/ICC: Used for washing and mild permeabilization of fixed cells to reduce nonspecific staining.

  • Immunohistochemistry (IHC): Washes tissue sections between antibody incubations to improve signal-to-noise.

  • Flow Cytometry: Employed as a wash buffer to limit nonspecific antibody interactions and cell clumping.

  • Magnetic Bead and Plate-Based Immunoassays: Washes beads and plates during capture and detection steps to reduce background.

  • Blocking Buffer Base: Combined with BSA or serum in PBS-T to block membranes, plates, and slides.

  • Phage Display Panning: Used in wash steps to decrease nonspecific binder carryover.

  • Microscopy Slide Preparation: Rinses fixed smears and slides to remove unbound probes or antibodies.

  • Microbial Immunostaining: Washes fixed Escherichia coli and Staphylococcus aureus samples during antibody labeling protocols.
