1X PBS, Dulbecco’s Solution without Calcium and Magnesium is a phosphate-buffered saline used for washing cells, sample handling, reagent preparation, and dilution steps where divalent cations are undesirable. It maintains physiological ionic strength and near-neutral pH and is commonly used with EDTA-based detachment protocols and for handling bacteria such as E. coli and mammalian cells.

  • Cell Culture Washing: Calcium- and magnesium-free isotonic rinse for removing serum and residual media without triggering adhesion or signaling.

  • Cell Detachment Workflows: Pre-rinse before trypsin/EDTA to enhance detachment efficiency and prevent cation-dependent adhesion.

  • Flow Cytometry: Washing and staining buffer that limits cell clumping by omitting divalent cations.
