Phosphate Buffer with 0.1% Tween 80 is an isotonic, pH-stabilized aqueous buffer containing a nonionic surfactant for wetting and dispersion during sample handling. The buffer maintains phosphate-based buffering capacity while the surfactant reduces surface tension to facilitate uniform suspension of particulates.

Applications include sample dilution, rinsing, and suspension of microorganisms in laboratory workflows, including handling of Escherichia coli, Staphylococcus aureus, and Salmonella enterica.

  • 0.778% Sodium Carbonate
  • 2.622% Monopotassium Phosphate
  • 0.1% Tween-80
  • Microbial Sample Diluent for Plate Counts: Used to suspend and dilute samples for CFU enumeration of Escherichia coli, Staphylococcus aureus, Salmonella enterica, Listeria monocytogenes, and Pseudomonas aeruginosa, with Tween 80 improving organism recovery from hydrophobic matrices.

  • Surface and Environmental Monitoring (Swab Rinse): Serves as a swab rinse/transport buffer, aiding detachment of microbes from surfaces and minimizing loss to plastics and swab fibers.

  • Neutralization/Recovery in Antimicrobial Efficacy Tests: Enhances recovery of stressed microorganisms after exposure to sanitizers or preservatives by reducing surfactant-related carryover and aiding dispersal.

  • Biofilm Disruption Prior to Enumeration: Facilitates dispersion of biofilms and clumps to yield more accurate plate counts and microscopy assessments.

  • Immunoassay Wash Steps (ELISA, Blotting): Used as a phosphate-based wash buffer with 0.1% Tween 80 to reduce nonspecific binding and improve wash efficiency.

  • Membrane Filtration Rinse Buffer: Employed to rinse filters and funnels during microbial load testing, reducing organism adhesion and surfactant residue on equipment.

  • Sample Preparation for Food/Cosmetic Microbiology: Extracts and disperses microorganisms from fatty, proteinaceous, or viscous products for downstream testing.

  • Resuspension and Transfer of Particulates: Maintains dispersion of cells and particles, reducing adsorption to tubes, pipette tips, and microfluidic channels.

  • General Buffering for Biochemical Procedures: Provides stable phosphate buffering with mild nonionic surfactant for workflows requiring reduced surface tension and consistent pH.

  • Device and Surface Rinses in Bioburden Testing: Used to rinse components and assemblies for quantitative recovery of microorganisms during validation and routine monitoring.

  • pH: 7.20 ± 0.50
