5X PBS-TBN Buffer is a concentrated phosphate-buffered saline containing a nonionic surfactant, a protein carrier, and an azide preservative. It is used to prepare working solutions for washing and blocking steps in immunoassays such as Western blotting, ELISA, and immunofluorescence. The buffer provides isotonic conditions for sample handling, incorporates gentle detergent for effective rinsing, includes a protein additive to minimize nonspecific interactions on assay surfaces, and contains a preservative to limit microbial contamination.

Dilute 1:5 with water to obtain a 1X working solution. Not recommended for applications involving horseradish peroxidase detection due to azide inhibition. Store at 2–8 °C; do not freeze.

  • 750mM Sodium Chloride
  • 250mM Sodium Phosphate Monobasic
  • 0.1% Tween-20
  • 0.5% Bovine Serum Albumin
  • 0.1% Sodium Azide

  • ELISA and Immunoassay Washing/Blocking: Reduces nonspecific binding during plate washes and blocking, improving signal-to-noise in antigen/antibody assays.

  • Western Blot Blocking and Antibody Incubation: Serves as a blocking and antibody diluent to minimize background on membranes.

  • Immunohistochemistry/Immunofluorescence: Used for tissue/cell blocking and antibody staining to reduce off-target interactions.

  • Flow Cytometry Staining/Wash Buffer: Maintains cell suspensions and minimizes nonspecific antibody binding during staining and washes.

  • Antibody Dilution and Short-Term Storage: Stabilizes antibodies with BSA and preserves solutions with sodium azide.

  • Bead-Based Immunoassays: Provides wash and blocking conditions for magnetic/agarose bead capture assays.

  • Immunoprecipitation/Co-IP: Used as a wash and dilution buffer to limit nonspecific protein–protein interactions.

  • Bio-Layer Interferometry/Sensor-Based Binding: Lowers nonspecific adsorption on sensors during kinetic assays.

  • Diagnostic Sample Handling: Preserves protein/antigen samples during processing; not intended for live cells due to sodium azide.

  • Microbial Antigen Detection Workflows: Employed in wash/block steps for assays targeting E. coli or Staphylococcus aureus antigens.
