1X PBS (phosphate-buffered saline) with 4 mM EDTA, pH 7.4, is an isotonic buffering solution for washing, dilution, and handling of cells and biomolecules. The EDTA component chelates divalent cations such as Ca2+ and Mg2+, reducing metal-dependent interactions.

Applications include nucleic acid workflows to limit nuclease activity, cell washing steps where calcium-dependent adhesion is minimized, and preparation or storage of samples where divalent cations are undesirable.

  • 4.0mM Ethylenediaminetetraacetic acid (EDTA)
  • Cell Washing and Resuspension: Isotonic buffer with EDTA that chelates divalent cations to reduce cell clumping and maintain cell integrity.

  • Enzyme-Free Cell Dissociation/Harvesting: EDTA disrupts Ca2+-dependent cell–cell and cell–matrix interactions to gently detach adherent cells.

  • Nuclease Protection in DNA/RNA Workflows: EDTA chelates Mg2+ to inhibit DNases, safeguarding nucleic acids during rinsing and handling.

  • Flow Cytometry: Used for staining and wash steps to minimize aggregate formation and improve single-cell suspensions.

  • Immunoassays (ELISA/IFA/Western Blot Washes): Provides stable pH and ionic strength while EDTA reduces metal-dependent nonspecific binding.

  • Protein and Enzyme Handling: Serves as a diluent/storage buffer; EDTA helps inhibit metalloproteases to preserve protein activity.

  • Magnetic Bead and Affinity Workflows: Minimizes cation-dependent nonspecific interactions during binding, washing, and elution steps.

  • Tissue and Slide Washing: Maintains physiological pH/tonicity for rinsing tissues and slides while removing divalent cations that can increase background.

  • Serum/Plasma Sample Preparation: EDTA chelation helps inhibit complement and metal-dependent enzymes during processing.
