1X Phosphate-Buffered Saline (PBS) with 4 mM EDTA, pH 7.4, is an isotonic, non-toxic buffer for routine laboratory use. The buffer maintains physiological pH and osmolarity during cell and tissue handling, washing steps, and sample preparation.

The inclusion of 4 mM EDTA provides chelation of divalent cations such as Ca2+ and Mg2+, reducing metal-dependent enzymatic activity and minimizing cell–cell and cell–matrix interactions during dissociation or washing procedures. Suitable for reagent preparation, sample dilution, and workflows requiring controlled ionic strength and pH 7.4.

  • 2.7mM Potassium Chloride
  • 4.3mM Disodium Phosphate
  • 1.4mM Monopotassium Phosphate
  • 4.0mM Ethylenediaminetetraacetic acid (EDTA)
  • Cell Washing and Suspension: Gentle isotonic buffer for washing and resuspending cells; EDTA chelates Ca2+/Mg2+ to reduce cell clumping.

  • Enzyme-Free Adherent Cell Detachment: Promotes dissociation of adherent cells by disrupting divalent cation–dependent cell–cell and cell–matrix interactions.

  • Flow Cytometry Sample Preparation: Maintains single-cell suspensions and improves sorting accuracy by minimizing aggregates.

  • Nuclease Inhibition in Nucleic Acid Workflows: EDTA chelates Mg2+/Ca2+ to inhibit DNases/RNases during sample washes and handling.

  • Complement Inactivation in Immunology Workflows: Prevents complement-mediated effects in serum/plasma handling and immunoassay wash steps.

  • Metal-Dependent Enzyme Quenching: Stops reactions that require divalent cations, aiding timed assay termination.

  • Biofilm and Bacterial Sample Processing: EDTA helps destabilize Gram-negative outer membranes to aid dispersion (e.g., Escherichia coli, Pseudomonas aeruginosa).
