Organic Sperm Wash Buffer, DNase Tested is a laboratory buffer for washing and processing sperm samples in molecular biology workflows. The formulation provides ionic strength, pH buffering, chelation, and an anionic surfactant to aid sample cleanup during preparation. Each lot is tested and shows no detectable DNase activity by an in vitro nuclease assay.

  • 50mM Sodium Chloride
  • 10mM Tris-HCl
  • 10mM Ethylenediaminetetraacetic acid (EDTA)
  • 2.0% SDS
  • Genomic DNA Extraction: SDS/EDTA lysis buffer for efficient isolation of high-molecular-weight DNA from cells and tissues.

  • Bacterial Cell Lysis: Disrupts cell walls and membranes to release nucleic acids from Escherichia coli, Staphylococcus aureus, and other bacteria.

  • Nuclease Inactivation: EDTA chelates divalent cations and SDS denatures proteins to inhibit DNases/RNases during nucleic acid purification.

  • Pre-Extraction Sample Preparation: Detergent-based lysis prior to phenol–chloroform or column-based DNA cleanup.

  • Protein Denaturation: SDS-driven unfolding and solubilization of proteins to reduce contaminants in DNA preparations.

  • Membrane Solubilization: Disrupts cellular and organelle membranes to improve nucleic acid yield from tough samples.

  • Southern Blot and Restriction Digest Prep: Generates clean, high-quality DNA suitable for downstream enzymatic workflows.

  • Microbial Genomics Workflows: Lysis and DNA prep from Gram-negative and Gram-positive organisms, including Escherichia coli and Bacillus subtilis.
