NZY Broth is a nutrient-rich medium formulated with an enzymatic digest of casein, yeast extract, sodium chloride, and a phosphate buffer. It supports the cultivation of Escherichia coli strains and is suitable for applications involving bacteriophage propagation and cloning workflows requiring vigorous bacterial growth in liquid culture.

The peptide and amino acid content from the casein digest provides nitrogen and growth factors, yeast extract supplies vitamins and nucleotides, sodium chloride maintains osmotic balance, and the phosphate component provides buffering capacity to help stabilize pH during incubation.

  • 1.00% NZ Amine
  • 0.50% Yeast Extract
  • 0.50% Sodium Chloride
  • 0.09% Potassium Phosphate Dibasic
  • Bacteriophage Propagation (M13/λ): Cultivation of host Escherichia coli for high-efficiency amplification of filamentous and lambda phage.

  • Plaque Assays and Top Agar Overlays: Base medium for preparing overlays and titrating phage on susceptible Escherichia coli strains.

  • Cloning and Plasmid Preparation: Growth of recombinant Escherichia coli for high-yield plasmid minipreps and midipreps.

  • Transformation Recovery: Post-transformation outgrowth of Escherichia coli to enhance viability and plasmid establishment.

  • Phage Display Library Amplification: Expansion and rescue of phagemid libraries using F+ Escherichia coli hosts.

  • Sanger Sequencing Workflows (M13-based): Preparation of phage templates and helper phage amplifications in Escherichia coli.

  • Competent Cell Quality Control: Benchmarking transformation efficiency using standard Escherichia coli strains.

  • General Rich Growth of Enteric Bacteria: Routine cultivation of laboratory Escherichia coli for subculturing and inoculum prep.
