Nematode Growth Media (NGM) Agar Plates are a defined solid medium used for culturing Caenorhabditis elegans and establishing bacterial lawns of E. coli strains such as OP50.

The medium includes buffering and ionic components for stable culture conditions, a peptone source for bacterial lawn formation, agar as the solidifying agent, and a cholesterol supplement required by nematodes.

  • 0.30% Sodium Chloride
  • 0.25% Tryptone
  • 2.00% Agar
  • 5µg/mL Cholesterol
  • 1mM Calcium Chloride
  • 1mM Magnesium Sulfate
  • 25mM Potassium Phosphate
  • C. elegans Culture and Maintenance: Routine propagation of C. elegans on lawns of Escherichia coli OP50 supported by 0.30% NaCl, 0.25% Tryptone, 2.00% Agar, and 5 µg/mL cholesterol.

  • Genetic Crosses and Strain Propagation: Setting up crosses, maintaining mutants, and isolating lines on standardized 60 mm plates.

  • RNAi by Feeding: Knockdown studies using Escherichia coli HT115(DE3) expressing dsRNA on buffered plates (25 mM potassium phosphate; 1 mM CaCl2, 1 mM MgSO4).

  • Developmental and Life-History Assays: Synchronization, growth rate, lifespan, and fecundity measurements under defined nutritional conditions.

  • Behavioral Assays: Chemotaxis, locomotion, egg-laying, and feeding (pharyngeal pumping) on reproducible agar surfaces.

  • Drug and Toxicology Screening: Plate-based exposure and dose–response testing for small molecules and environmental toxins.

  • Host–Pathogen Interaction Studies: Infection assays with Pseudomonas aeruginosa PA14, Serratia marcescens, or Enterococcus faecalis to assess innate immunity.
