Middlebrook 7H10 Media Agar Plates are solid culture media prepared from Middlebrook 7H10 base supplemented with glycerol and OADC enrichment. The medium provides defined nutrients, fatty acids, vitamins, and cofactors required for the cultivation of mycobacteria.

The plates support colony development of members of the Mycobacterium tuberculosis complex and non-tuberculous mycobacteria, including fast-growing species such as Mycobacterium smegmatis. Glycerol serves as a carbon source for glycerol-utilizing strains, and OADC supplies albumin, dextrose, catalase, and oleic acid to stabilize fatty acids and mitigate peroxide effects. Suitable for streaking, colony isolation, and routine maintenance of mycobacterial cultures under appropriate biosafety conditions.

  • 1.9% 7H10 Agar Medium
  • 0.5% Glycerol
  • 10.0% OADC Enrichment Media
  • Isolation and Cultivation of Mycobacteria: Supports growth and isolation of Mycobacterium tuberculosis and non-tuberculous mycobacteria such as Mycobacterium avium from clinical and environmental specimens.

  • Colony Forming Unit (CFU) Enumeration: Used for plating and counting mycobacterial CFUs in infection models, growth kinetics, and drug studies.

  • Antimicrobial Susceptibility Testing: Employed for agar proportion and MIC testing of mycobacteria, including Mycobacterium tuberculosis complex.

  • Maintenance and Subculture: Routine propagation and maintenance of laboratory strains such as Mycobacterium smegmatis and Mycobacterium tuberculosis.

  • Post-Decontamination Recovery: Recovery and purification of mycobacteria after specimen decontamination (e.g., NALC-NaOH processing).

  • Genetic Selection and Screening: Plating of transformants and mutant libraries with selective antibiotics for mycobacterial genetics.

  • Colony Morphology and Pigment Assessment: Evaluation of colony characteristics and pigment production for species characterization.

  • Non-tuberculous Mycobacteria (NTM) Studies: Isolation and quantification of NTM, including Mycobacterium avium complex, from clinical and environmental sources.
