Mueller Hinton II Cation Adjusted Agar Plates are prepared from a defined cation-adjusted nutrient base solidified with agar to provide a consistent surface for antimicrobial susceptibility testing of aerobic, nonfastidious bacteria. The cation content is controlled to specified calcium and magnesium levels to support standardized test conditions.

This medium is commonly used for disk diffusion and dilution-based methods as described in recognized standards. It supports the growth of organisms such as Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Enterococcus faecalis, and members of the Enterobacteriaceae.

  • 2.2% Mueller Hinton II Broth (Cation-Adjusted)
  • 1.5% Agar
  • Antimicrobial Susceptibility Testing (Kirby–Bauer Disk Diffusion): Standardized cation-adjusted 2.2% Mueller Hinton II Broth, 1.5% agar plates for zone diameter testing of nonfastidious aerobes such as Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa.

  • MIC Determination with Gradient Diffusion Strips: Supports Etest/M.I.C.E.-type assays to determine minimum inhibitory concentrations against organisms like Acinetobacter baumannii and Klebsiella pneumoniae.

  • Quality Control of Antimicrobial Disks and Media: Verification of disk potency and media performance using reference strains on cation-adjusted Mueller Hinton II agar.

  • Routine Cultivation and Inoculum Preparation: Growth of nonfastidious bacteria and preparation of standardized inocula (e.g., 0.5 McFarland) for downstream AST.

  • AST of Fastidious Organisms with Supplements: Used with appropriate additives (e.g., 5% sheep blood for Streptococcus pneumoniae, HTM supplement for Haemophilus influenzae) for standardized testing.

  • Purity Checks and Colony Isolation: Ensures pure cultures and even lawns prior to ID and susceptibility workflows.

  • Antibiotic Screening in Research: Evaluation of novel antimicrobial compounds and combinations against targets such as Enterococcus faecalis and Pseudomonas aeruginosa.
