Modified 2X M9 Salts Media is a defined minimal medium for cultivation and maintenance of E. coli and other enteric bacteria. It provides a balanced inorganic salt base at 2X strength, a glucose carbon source, and essential vitamin support for routine growth and propagation in minimal conditions.

The formulation includes divalent cations for membrane stability and enzyme function, thiamine for cofactor-dependent metabolism, and dextrose to support central carbon metabolism. Suitable for preparing liquid cultures or as a base for agar plates when supplemented with an appropriate gelling agent.

  • 2X M9 Salts
  • 2mM Magnesium Sulfate
  • 0.2mM Calcium Chloride
  • 1µg/mL Thiamine
  • 2% Cerelose Anhydrous Dextrose (Glucose)
  • Defined Minimal Growth of Escherichia coli: Culturing and maintenance of Escherichia coli and other enteric bacteria in a strictly defined medium with 2X M9 Salts, 2% glucose, thiamine, Mg2+, and Ca2+.

  • Auxotroph/Prototroph Selection: Screening genetic mutations using a defined carbon source (2% glucose) and thiamine (1 µg/mL) to assess biosynthetic requirements.

  • Plasmid Maintenance and Expression Studies: Controlled growth conditions for antibiotic selection and recombinant protein expression with reduced background metabolites.
