5X M9 Minimal Media Salts is a concentrated inorganic salt base for preparing M9 minimal medium. The formulation provides a phosphate buffering system with sodium and potassium ions, sodium chloride for ionic balance, and ammonium as an inorganic nitrogen source. When supplemented with a suitable carbon source and divalent cations, it is used to cultivate E. coli and other non-fastidious bacteria under defined nutrient conditions and for applications requiring controlled media composition.

Intended for dilution to working strength and supplementation with a carbon source and magnesium and calcium salts prior to use. Optional additions such as vitamins, amino acids, or antibiotics may be included according to experimental requirements.

  • 2.98% Disodium Phosphate
  • 1.63% Monopotassium Phosphate
  • 0.26% Sodium Chloride
  • 0.51% Ammonium Chloride
  • Defined Bacterial Culture: Preparation of M9 minimal medium for growth of Escherichia coli and other bacteria under strictly defined nutrient conditions.

  • Auxotroph and Mutant Selection: Screening and selection of metabolic auxotrophs by omitting specific nutrients or amino acids.

  • Carbon Source Utilization Studies: Assessment of substrate preference and catabolite repression by supplementing with glucose, glycerol, acetate, or other carbon sources.

  • Isotopic Labeling: 13C/15N labeling of proteins and metabolites for NMR, MS, and fluxomics in Escherichia coli.

  • Recombinant Protein Expression: Expression in Escherichia coli using defined media to reduce background metabolites and enable label incorporation.

  • Chemostat and Continuous Culture: Controlled nutrient-limited growth for physiology and evolution experiments.

  • Transport and Uptake Assays: Measurement of nutrient transport kinetics under defined ionic and pH conditions.

  • Antibiotic Selection and Plasmid Maintenance: Defined growth conditions to standardize growth rates and selection pressure.

  • Gene Regulation and Induction Studies: Use with specific inducers (e.g., arabinose, lactose) to analyze promoter control in minimal background.

  • Metabolic Engineering: Tuning of nitrogen, phosphorus, and carbon availability to optimize pathway flux.

  • Microscopy and Fluorescence Assays: Low-background, low-autofluorescence medium for imaging of bacterial reporters.

  • Comparative Physiology Across Species: Defined-media growth comparisons for strains of Escherichia coli, Pseudomonas aeruginosa, and related bacteria.
