M9 Minimal Salts Agar Plates with 1% Glucose, Proline-250 is a defined minimal agar medium for the cultivation of E. coli and other bacteria under chemically defined conditions. Glucose serves as the sole carbon and energy source, thiamine supplies a required vitamin cofactor, and magnesium and calcium provide essential divalent cations. Proline is included as a defined amino acid supplement for work with proline-auxotrophic strains on solid medium.

  • 1X M9 Salts
  • 1% Glucose
  • 0.1mM Calcium Chloride
  • 1.0mM Thiamine
  • 0.2mM Magnesium Sulfate
  • 250µg/mL Proline
  • 1.5% Agar
  • Selection of Auxotrophs/Prototrophs: Isolation and screening of amino acid biosynthesis mutants, including proline auxotrophs and complementation using 250 µg/mL proline.

  • Defined Carbon Source Utilization: Assessment of metabolic ability to grow on 1% glucose in a minimal, defined background (1X M9 salts).
